Cecal ligation and puncture induced sepsis

چکیده
Purpose: Multiresistant and vancomycin resistant Enterococcus faecium (VRE) can cause serious infections in hospitalized patients with various co-morbid diseases. We investigated the course of VRE peritonitis after cecal ligation and puncture (CLP)-induced sepsis and compared this to sham operated mice. Methods: Mice were subjected to CLP or sham surgery. Forty-eight hours thereafter four groups were created by subjecting mice to peritoneal injection of either VRE or saline. Results: Mice infected with VRE after CLP were severely impaired in eliminating VRE from the peritoneal cavity and distant body sites. These mice failed to mount an early inflammatory response at the primary site of VRE infection. VRE superinfection did not influence CLPinduced organ damage or polymicrobial bacterial loads. Conclusions: Sublethal polymicrobial sepsis greatly facilitates infection and dissemination of VRE. VRE does not influence the course of CLPinduced sepsis.

A New Approach to Monitoring and Evaluation of Cecal Ligation and Puncture Sepsis Model
Background: Sepsis is a systemic inflammatory disease in response to the pathogens that leads to vital organ failures the failure of vital organs. Appropriate animal models should be developed to measure the effectiveness of therapeutic methods. Cecal Ligation and Puncture (CLP) is the most widely used methods of creating the sepsis model. Some variables interfere in the creation of the CLP mod...
متن کاملMedian lethal needle caliber in two models of experimental sepsis.
PURPOSE To estimate the median lethal needle caliber (LC50) of a new experimental sepsis model and compare it to the LC50 of the cecal ligation and puncture (CLP) sepsis model. METHODS Male albino Wistar rats were studied (n=22). Animals were allocated into two study groups. In Group I, experimental sepsis was induced by cecal ligation and puncture. In Group II, experimental sepsis was induce...
متن کاملValidation of a Murinic Model of Sepsis Induced by Cecal Ligation and Puncture Technique
Sepsis is considered to be a systemic response to an infection. This pathology is an common cause for mortality and morbidity being the primary cause of death among patients in the Intensive Care Units. Because of the increasing need for a standardised animal model of sepsis we decided to validate a murinic one using the cecal ligation and puncture (CLP) technique and easy to obtain physiologic...
متن کاملSepsis modeling in mice: ligation length is a major severity factor in cecal ligation and puncture
BACKGROUND The cecal ligation and puncture (CLP) model, a gold standard in sepsis research, is associated with an important variability in mortality. While the number of punctures and needle size is well described in CLP animal studies, the length of cecal ligation is often not. The relationship between cecal ligation and survival in mice is briefly reported in the literature; therefore, we dev...
متن کاملTransient receptor potential melastatin 2 protects mice against polymicrobial sepsis by enhancing bacterial clearance.
BACKGROUND Recent studies suggest that the transient receptor potential melastatin 2 (TRPM2) channel plays an important role in inflammation and immune response. However, the role and mechanism of TRPM2 in polymicrobial sepsis remain unclear. METHODS The authors explored the effects of genetic disruption of TRPM2 on mortality (n = 15), bacterial clearance (n = 6), organ injury, and systemic i...
